Adoptability
Although high-throughput screening (HTS) is used extensively to streamline and scale-up drug screening through automated systems, it relies heavily on conventional 2D cell culture platforms, which poorly mimic in vivo conditions. Despite their advantages over 2D systems in recapitulating the tumor microenvironment, many organ-on-a-chip platforms are not compatible with high content screening (HCS) systems and often require custom equipment for routine screening procedures. Further, many platforms lack the ability to be adopted to conventional HTS and HCS systems.
Santiagosic’s microfluidic 96-Curio platform incorporates a high-throughput SBS format which is compatible with automated fluidic handlers, robotic systems, and HCS. These key features make the Curiochips Microphysiological Systems (MPS) highly adoptable to any standard instruments for high-throughput screening.

Robust High-Throughput Reproducibility
Conventional 2D tissue cultures as well as animal testing are often incapable of recapitulating human pathophysiology. This can lead to discrepancies between preclinical and clinical studies resulting in costly high failure rates during the drug discovery process.
Commercially available microfluidic Organ-on-a-Chip platforms allow researchers to better model human diseases. Thus, in theory they are capable of leading to a more robust and meaningful drug discovery process. However, many current Organ-on-a-Chip and Microphysiological Systems (MPS) are lacking in reproducibility due to their difficulty in use and unpredictable gel patterning.

Our Organ-on-a-Chip microfluidic screening platforms (Curiochips) incorporate a novel microfluidic design which integrates patterned hydrogels or ECM components with Organ-on-a-chip 3D co-cultures. This makes it possible to perform AI-based data image analysis, automated data normalization, data management, image generation and data processing in a reproducible manner.

The large quantity and high complexity of the data generated by high-content imaging technology preclude visual or manual analysis and require advanced computational methods to fully explore the wealth of information and accomplish high-throughput processing.
Our deep learning-based computational algorithm improves the accuracy and sensitivity of currently available image analysis. Its automated analysis enables high-throughput screening by removing human intervention, therefore making the analysis result more objective and reproducible. Furthermore, it can identify subtle and complex morphological features which may be unnoticeable or in discriminable to the human eye, and then quantify them into continuous scores.
